Column chromatography is a well-establishedbioprocessing purification method. Ion-exchangeresin columns have been especially effective andreliable parts of successful purification. Affinity, hydrophobic interaction and other media effect highresolution purifications of specific target molecules.

But column-packing and packing validation can betime-consuming and expensive. The high cost ofprocess columns, along with the cost of associatedequipment, cleaning, and storage validation canimpact process economics. Processing biomolecules for initial capture of targetproduct, purification of the target molecule, and the removal of potentially harmful contaminants fromthe purified product, all place different demands on the chromatography process. Membrane ChromatographyMustang® membranes are an innovativeion-exchange support containing functionalquaternary amine (Q) or sulfonic acid (S) groups. The large pores of Mustang membranes allow biomolecules to access all the binding sites bydirect fluid convection, rather than being dependenton the restrictions imposed by diffusive flow, themethod of operation of chromatography resins. Thisresults in high dynamic flow, very steep breakthroughcurves, equivalent or higher capacity thanresins for most target species, and process flowrates that can be as high as 25 to 50 columnvolumes per minute.

Mustang ion-exchange chromatography capsulesare prepacked, single-use systems specificallydesigned to provide high throughputs. Therefore, nocolumn packing or cleaning is required, resulting in significantly shorter processes cycles. Because of thehigh dynamic binding capacities of Mustangcapsules, they are particularly well-suited for use inthe initial purification of the target molecule, or forremoving contaminating molecules, such as DNA orviruses, in the final polishing steps. Mustang E isused to remove endotoxin from water, saline, andbuffer solutions.
